Biological Activity

JSH-23 is an inhibitor of NF-κB transcriptional activity that inhibits LPS-stimulated NF-κB transcriptional activity with an IC50 of 7.1 μM in RAW 264.7 macrophages. Mechanistically, JSH-23 selectively interferes with LPS-induced NF-κB nuclear translocation without affecting IκB degradation, thereby inhibiting downstream pro-inflammatory gene expression and inflammatory phenotypes mediated by the NF-κB signaling pathway. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
