Biological Activity

Onametostat (JNJ-64619178) is a potent and selective protein arginine methyltransferase 5 (PRMT5) inhibitor targeting the PRMT5-MEP50 complex. By suppressing PRMT5-mediated symmetric dimethylation of arginine residues on substrate proteins, Onametostat disrupts RNA splicing machinery and downstream oncogenic signaling pathways. This targeted inhibition effectively impairs tumor cell proliferation and induces apoptosis in PRMT5-dependent cancer models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
