Biological Activity

JK184 is a potent Hedgehog (Hh) signaling pathway inhibitor that targets Gli transcriptional activity with an IC50 of 30 nM in mammalian cells. By suppressing Gli-mediated gene transcription downstream of Smoothened, JK184 blocks Hedgehog pathway activation, leading to decreased expression of target genes involved in cell proliferation and survival. Consequently, inhibition of this cascade attenuates the growth of Hedgehog-dependent tumor cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
