Biological Activity

JHU-083 is a novel prodrug of 6-diazo-5-oxo-L-norleucine (DON) that acts as a glutamine antagonist and selectively blocks glutaminase activity. By inhibiting glutamine metabolism, JHU-083 disrupts nutrient flux, suppresses tumor cell proliferation, and reprograms effector T cell metabolism to enhance antitumor immune responses. Additionally, JHU-083 modulates neuroinflammatory signaling in brain CD11b+ cells to alleviate stress-induced behavioral deficits. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
