INH1 is a cell-permeable Hec1 inhibitor that specifically disrupts the interaction between highly expressed in cancer protein 1 (Hec1) and NIMA-related kinase 2 (Nek2). By disrupting the Hec1/Nek2 complex, INH1 impairs kinetochore function during mitosis, leading to chromosome missegregation, spindle assembly activation, and cellular arrest. This downstream mitotic disruption effectively inhibits cancer cell proliferation and suppresses tumor growth in cellular and preclinical models.
