GDC-0077 (Inavolisib) is a potent and selective inhibitor of PI3Kα. It competes with ATP for binding to PI3Kα, thereby inhibiting the phosphorylation of PIP2 to PIP3. Blockade of this enzymatic activity suppresses downstream AKT/mTOR signaling, leading to reduced cellular proliferation and the induction of apoptosis in PI3Kα-dependent cancer cells.[1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
