IDH-305 is an orally available, mutant-selective, and brain-penetrant inhibitor of isocitrate dehydrogenase 1 (IDH1) that targets IDH1 R132H, IDH1 R132C, and wild-type IDH1. By inhibiting mutant IDH1, it blocks the production of the oncometabolite 2-hydroxyglutarate (2-HG), suppresses downstream oncogenic pathways, and promotes cellular differentiation in IDH1-mutated cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
