Biological Activity

Samuraciclib (ICEC0942) hydrochloride is an orally bioavailable cyclin-dependent kinase 7 (CDK7) inhibitor with an IC50 of 40 nM, displaying 45-, 15-, 230-, and 30-fold selectivity over CDK1, CDK2, CDK5, and CDK9, respectively. By inhibiting CDK7-mediated phosphorylation of RNA polymerase II and cell cycle CDKs, it suppresses gene transcription and cell cycle progression. Consequently, samuraciclib blocks cell proliferation and induces cell cycle arrest and apoptosis in cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
