H3B-6527 is a selective covalent inhibitor of FGFR4 (IC50 <1.2 nM), displaying at least 250-fold selectivity over FGFR1 (IC50 320 nM), FGFR2 (IC50 1290 nM), and FGFR3 (IC50 1060 nM). By selectively targeting FGFR4, H3B-6527 blocks FGF19-driven FGFR4 signaling and suppresses downstream ERK phosphorylation. This inhibition impairs cell proliferation and tumor growth in hepatocellular carcinoma models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
