Biological Activity

GW441756 is a potent and selective inhibitor of TrkA kinase (IC50 2 nM) with minimal inhibitory activity against c-Raf1 and CDK2. By selectively inhibiting cellular TrkA-mediated signaling pathways, GW441756 suppresses downstream neurotrophin signaling and promotes a marked increase in caspase-3 expression, ultimately triggering apoptotic cell death in target cancer cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
