Guanabenz is an orally active central α2-adrenergic receptor agonist and a selective GADD34 (PPP1R15A) inhibitor. By selectively binding GADD34, Guanabenz inhibits the protein phosphatase 1 (PP1) complex to prevent the dephosphorylation of eukaryotic translation initiation factor 2 subunit alpha (eIF2α). This sustained eIF2α phosphorylation attenuates global protein synthesis and protects cells against endoplasmic reticulum (ER) stress-induced proteotoxicity. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
