Biological Activity

GSK4112 is a functional nuclear receptor Rev-erbα agonist with an EC50 of 0.4 μM. Upon binding to Rev-erbα, GSK4112 recruits co-repressors to modulate circadian clock-regulated gene networks and repress the transcription of key gluconeogenic genes in liver cells. This downstream transcriptional repression subsequently reduces hepatic glucose output in primary hepatocytes, linking nuclear receptor activity to metabolic gene regulation.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
