GSK343 is a potent, selective histone methyltransferase EZH2 inhibitor with 60-fold selectivity over EZH1 and greater than 1000-fold selectivity over other histone methyltransferases. By targeting the catalytic subunit of the PRC2 complex, it inhibits trimethylation of lysine 27 on histone H3 (H3K27me3) to regulate gene expression, decrease cell proliferation, and induce autophagy in cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
