GSK269962A HCl is a potent and selective Rho-associated protein kinase (ROCK) inhibitor, targeting ROCK1 (IC50 1.6 nM) and ROCK2 (IC50 4 nM). By impairing ROCK-mediated kinase activity within the ROCK pathway, this compound suppresses downstream substrate phosphorylation required for actin cytoskeletal reorganization and smooth muscle contraction, thereby inhibiting cell migration, reducing vascular tone, and suppressing pathological cell proliferation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
