GS-621763 is an orally bioavailable prodrug of GS-441524 that targets viral RNA-dependent RNA polymerase (RdRp). It exhibits potent antiviral activity against SARS-CoV-2 and Middle East respiratory syndrome coronavirus (MERS-CoV) in human primary lung cell cultures. Blockade of viral RdRp suppresses viral RNA replication, thereby attenuating viral load and protecting against severe lung pathology in vivo. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
