GLPG3312 is a potent pan-salt-inducible kinase (SIK) inhibitor targeting SIK1, SIK2, and SIK3. By suppressing SIK activity, the compound modulates CREB-mediated transcription and downregulates pro-inflammatory signaling pathways. This inhibition promotes immunoregulatory gene expression, leading to increased interleukin-10 production and suppressed secretion of pro-inflammatory cytokines in primary myeloid cells, demonstrating potent anti-inflammatory and immunoregulatory functions in vitro and in vivo. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
