Givinostat (ITF-2357) is an orally active histone deacetylase (HDAC) inhibitor targeting class I and class II HDACs. HDAC inhibition alters gene transcription and attenuates downstream inflammatory signaling pathways. In peripheral blood mononuclear cells, Givinostat suppresses the production of pro-inflammatory cytokines, including TNF-α, IL-1β, IL-6, and IFN-γ. Additionally, treatment inhibits total intracellular IL-1β synthesis at concentrations below 25 nM and promotes apoptotic cell death in hematological cells. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
