Biological Activity

Divarasib (GDC-6036) is an orally available, potent covalent inhibitor of KRAS G12C that irreversibly binds to the switch II pocket of the inactive GDP-bound protein, thereby blocking GTP binding and oncogenic activation. Target inhibition impairs downstream MAPK signaling through the RAF/MEK/ERK pathway, resulting in suppressed cellular proliferation and robust tumor growth inhibition across multiple KRAS G12C-positive cancer cell lines and models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
