Gambogic acid is a natural apoptosis inducer and competitive inhibitor of anti-apoptotic Bcl-2 family proteins, including Bcl-XL, Bcl-2, Bcl-W, Bcl-B, Bfl-1, and Mcl-1, while also activating caspases. By binding to target proteins such as the transferrin receptor and modulating the mitochondrial apoptotic pathway, gambogic acid triggers caspase cascade activation, suppresses cell proliferation, and induces apoptosis in cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
