Biological Activity

FLLL32 is a potent small-molecule JAK2 and STAT3 inhibitor that suppresses STAT3 activation. By blocking STAT3 signaling, it effectively inhibits IL-6- and IFNα-stimulated STAT3 phosphorylation at Tyr705. This targeted inhibition disrupts downstream signaling cascades, downregulating anti-apoptotic proteins such as Bcl-2 and survivin, suppressing cell proliferation, and inducing apoptosis in various cancer cell lines including breast and pancreatic cancer cells. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
