FDI-6 is a small-molecule inhibitor of the Forkhead box protein M1 (FOXM1) transcription factor. It directly binds to FOXM1 and displaces it from genomic DNA, suppressing FOXM1-mediated transcriptional activity. By inhibiting this oncogenic transcriptional network, FDI-6 downregulates essential cell-cycle regulators, leading to cell cycle arrest and suppressing the proliferation of various human cancer cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
