Erastin is a potent system Xc- inhibitor and ferroptosis inducer. It affects Nrf2/HO-1 and AMPK signaling pathways, by depleting intracellular glutathione, decreasing GPX4 activity, and promoting lipid peroxidation and ROS accumulation, to induce ferroptosis and inhibit cancer cell proliferation.
