Biological Activity

Olodanrigan (EMA401) is an orally active, highly selective, peripherally restricted antagonist of the angiotensin II type 2 receptor (AT2R). By blocking AT2R signaling, olodanrigan inhibits angiotensin II-induced activation of p38 and p42/p44 MAPK pathways. This inhibition suppresses hyperexcitability and neurite sprouting in dorsal root ganglion neurons, ultimately mediating its targeted analgesic activity in preclinical pain models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
