Biological Activity

DNPH1i (N6-benzyladenosine) is a purine nucleoside derivative functioning as a competitive inhibitor of 2′-deoxynucleotide 5′-phosphate N-hydrolase 1 (DNPH1) and adenosine deaminase. By blocking DNPH1-mediated hydrolysis of cytotoxic 5-hydroxymethyl-2′-deoxyuridine monophosphate, it leads to genomic integration of non-canonical nucleotides during DNA replication. This disruption of nucleotide pool maintenance potentiates poly(ADP-ribose) polymerase inhibitor sensitivity and induces synthetic lethality in BRCA-deficient cells. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
