Diacerein is an anthraquinone inhibitor of pro-inflammatory cytokine interleukin-1β (IL-1β) production and signaling. By downregulating IL-1β expression and blocking downstream activation of NF-κB signaling, diacerein suppresses the production of matrix metalloproteinases, specifically MMP-1 and MMP-3. Consequently, this inhibition attenuates cartilage extracellular matrix degradation, reduces chondrocyte apoptosis, and mitigates tissue inflammation associated with arthritic pathologies. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
