Derazantinib is an orally bioavailable pan-FGFR inhibitor targeting FGFR1 (IC50 4.5 nM), FGFR2 (IC50 1.8 nM), FGFR3 (IC50 4.5 nM), and FGFR4 (IC50 34 nM), with additional inhibitory activity against RET, DDR2, PDGFRβ, VEGFR, and KIT. By inhibiting FGFR autophosphorylation, derazantinib blocks downstream AKT/mTOR and ERK/MAPK signaling pathways, leading to suppressed cell proliferation and induced apoptosis in FGFR-dependent cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
