Biological Activity

Zedoresertib (Debio-0123) is a potent small-molecule inhibitor targeting WEE1 kinase. By inhibiting WEE1 activity, it prevents the inhibitory phosphorylation of CDC2 (CDK1), thereby abrogating the G2 cell cycle checkpoint. This loss of checkpoint control forces cells with DNA damage to prematurely enter mitosis, leading to severe genomic instability, persistent double-strand DNA breaks, and cell death via apoptosis within cancer cells dependent on the DNA damage response pathway. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
