Biological Activity

Dasabuvir (ABT-333) is a non-nucleoside inhibitor of HCV NS5B RNA-dependent RNA polymerase, inhibiting recombinant NS5B polymerases from HCV genotype 1a and 1b isolates with IC50 values between 2.2 and 10.7 nM. By targeting the viral NS5B polymerase, it disrupts viral genome replication pathways and reduces viral RNA synthesis. This mechanism displays over 7,000-fold selectivity over human polymerases, suppressing hepatitis C virus replication in replicon systems. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
