Biological Activity

Darunavir (TMC114) is a potent nonpeptidic HIV protease inhibitor that selectively targets HIV-1 protease. By binding to the active site of the viral protease, it blocks the cleavage of Gag-Pol polyproteins into functional structural components and essential viral enzymes. This inhibition prevents viral maturation, resulting in the release of non-infectious immature virus particles and effective suppression of viral replication. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
