Daprodustat (GSK1278863) is an orally active hypoxia-inducible factor-prolyl hydroxylase (HIF-PH) inhibitor. By inhibiting HIF-PH enzymes, it prevents the proline hydroxylation and subsequent ubiquitin-proteasomal degradation of hypoxia-inducible factor alpha (HIF-α) subunits. This leads to the stabilization and nuclear accumulation of HIF-1α and HIF-2α, activating transcription of target genes in the HIF signaling pathway that induce erythropoietin expression and promote erythropoiesis. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
