Dapivirine (TMC120) is a potent non-nucleoside reverse transcriptase inhibitor (NNRTI) targeting HIV-1 reverse transcriptase. By binding to a hydrophobic pocket adjacent to the active site of viral reverse transcriptase, it inhibits RNA-dependent DNA synthesis and blocks viral replication. Dapivirine demonstrates broad-spectrum activity against diverse HIV-1 isolates, including various NNRTI-resistant strains, effectively suppressing viral infection and propagation in host cells. [1]
