Biological Activity

Danicopan is a potent, selective, and orally active small-molecule inhibitor targeting factor D with a Kd of 0.54 nM for human factor D. By reversibly binding factor D, it blocks the catalytic cleavage of factor B and prevents the formation of alternative pathway C3 convertase. This inhibition suppresses downstream alternative pathway of complement activity, preventing complement-mediated hemolysis and membrane attack complex formation. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
