Biological Activity

Dabrafenib (GSK2118436) is a selective inhibitor of mutant BRAF V600E (IC50 0.7 nM), displaying 7-fold less potency against wild-type BRAF and 9-fold less potency against C-RAF. By inhibiting oncogenic BRAF kinase activity, it suppresses downstream MAPK/ERK pathway signaling, thereby decreasing cancer cell proliferation and inducing cell cycle arrest and apoptosis in BRAF-mutant melanoma models. A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
