Dabigatran etexilate mesylate is an orally active prodrug of dabigatran, a selective and reversible direct thrombin inhibitor. Its active moiety, dabigatran, selectively inhibits human thrombin with a Ki of 4.5 nM and inhibits thrombin-induced platelet aggregation with an IC50 of 10 nM. By directly targeting free and clot-bound thrombin, dabigatran prevents the cleavage of fibrinogen into fibrin, effectively suppressing downstream blood coagulation cascades and platelet activation phenotypes. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
