Cyclosporin A is an immunosuppressive cyclic peptide that binds cyclophilin to inhibit the serine/threonine phosphatase calcineurin. By blocking calcineurin activity, Cyclosporin A prevents the dephosphorylation and nuclear translocation of the transcription factor NFAT. This blockade disrupts T-cell receptor-mediated signaling pathways, inhibiting interleukin-2 (IL-2) transcription and suppressing downstream T-lymphocyte activation and proliferation. [1][2]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
