Biological Activity

CU-CPT9a is a potent and specific Toll-like receptor 8 (TLR8) antagonist with an IC50 of 0.5 nM. By binding to the TLR8 dimer interface, CU-CPT9a inhibits agonist-induced conformational changes, effectively blocking downstream NF-kB signaling pathways. Consequently, this targeted inhibition suppresses TLR8-mediated proinflammatory cytokine production, including TNF-α and IL-1β, in human primary immune cells. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
