Biological Activity

Pelabresib (CPI-0610) is a potent and selective benzoisoxazoloazepine inhibitor of BET bromodomains, including BRD4. By selectively binding to BET bromodomains, pelabresib disrupts the recruitment of bromodomain proteins to chromatin, thereby suppressing downstream gene expression, including MYC and NSD3 target genes. This inhibition disrupts transcriptional programs involved in oncogenesis and cellular proliferation, leading to suppressed tumor cell growth and induced apoptosis in hematological tumor models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
