Ciliobrevin D is a cell-permeable, reversible, and selective antagonist of the AAA+ ATPase cytoplasmic dynein. By inhibiting dynein motor activity, it impairs intracellular transport along microtubules, leading to the perturbation of primary cilia formation and suppression of Hedgehog (Hh) signaling. Additionally, Ciliobrevin D disrupts mitotic spindle assembly and chromosome alignment, offering a valuable tool to probe dynein-dependent cellular processes. [1]
