Ciclesonide is a synthetic glucocorticoid receptor agonist that functions as an inhaled prodrug for treating obstructive airway diseases. Upon enzymatic cleavage in lung tissue, it yields its active metabolite desisobutyryl-ciclesonide, which binds the glucocorticoid receptor with high affinity. Activation of the receptor inhibits NF-kB pathway signaling and downregulates pro-inflammatory cytokine secretion, ultimately suppressing inflammatory cellular infiltration and mucosal hyperresponsiveness. - Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
