Biological Activity

Chloroquine diphosphate is an antimalarial and antirheumatoid 4-aminoquinoline derivative that acts as an activator of ataxia telangiectasia mutated (ATM) kinase and an inhibitor of toll-like receptor (TLR) signaling. By increasing endosomal pH and inhibiting lysosomal degradation, chloroquine impairs autophagic flux and blocks endosomal TLR activation. Additionally, chloroquine induces ATM autophosphorylation and downstream signaling independently of direct DNA double-strand break induction, modulating cellular stress responses and immune activation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
