CFSE (CFDA-SE) is a cell-permeable fluorescent staining dye widely used to track cell proliferation and motility. Upon entering cells, cleavage by intracellular esterases yields amine-reactive fluorescein derivatives that covalently couple via succinimidyl groups to intracellular lysine residues and proteins. The resulting fluorescent conjugates are stably retained within living cells and partitioned equally between daughter cells during division, enabling tracking of cell division kinetics by flow cytometry. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
