Delanzomib (CEP-18770) is an orally active inhibitor of proteasome chymotrypsin-like activity with an IC50 of. It disrupts the ubiquitin-proteasome pathway, preventing IkB degradation and suppressing downstream NF-kB signaling. This inhibition causes accumulation of ubiquitinated proteins, downregulates anti-apoptotic factors such as XIAP, and effectively inhibits tumor cell proliferation while inducing apoptosis in multiple myeloma and solid tumor cells.
