Capsazepine is a synthetic compound that acts as a competitive antagonist of the transient receptor potential vanilloid 1 (TRPV1) channel. It selectively inhibits capsaicin-induced channel activation, thereby suppressing intracellular calcium influx and the subsequent depolarization of primary sensory neurons. By attenuating TRPV1-mediated nociceptive pathway signaling, capsazepine blocks capsaicin-evoked membrane currents, cellular excitation, and nociceptive responses in rodent pain models. [1]
