Emavusertib (CA-4948) is an orally bioavailable dual inhibitor targeting interleukin-1 receptor-associated kinase 4 (IRAK4) and FMS-like tyrosine kinase 3 (FLT3). Suppressing IRAK4 disrupts Toll-like receptor and interleukin-1 receptor signaling pathways, impairing downstream myddosome assembly and NF-kB activation. This target blockade downregulates pro-inflammatory signaling and suppresses leukemic cell growth in hematologic malignancies such as acute myeloid leukemia.
