BV6 is a bivalent SMAC mimetic and dual antagonist of cellular inhibitor of apoptosis proteins (cIAP1/cIAP2) and X-linked inhibitor of apoptosis protein (XIAP). By inducing rapid autoubiquitination and proteasomal degradation of cIAPs, BV6 stimulates non-canonical NF-κB signaling and promotes autocrine TNFα production. This pathway activation leads to the assembly of a pro-apoptotic receptor-interacting protein 1 (RIP1)-dependent complex, ultimately driving caspase cascade activation and apoptotic cell death in susceptible cancer cells. - Preheat media/saline to 37°C.
