Biological Activity

BRM/BRG1 ATP Inhibitor-1 (BRM is an orally active, allosteric dual inhibitor of Brahma Homolog (BRM)/SMARCA2 and Brahma-related gene 1 (BRG1)/SMARCA4 ATPase activity. By disrupting the catalytic ATPases of the SWI/SNF chromatin remodeling complex, the compound suppresses BRM-dependent transcriptional regulation, leading to downstream antiproliferative responses and significant tumor growth inhibition in BRG1-mutant cancer xenograft models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
