Biological Activity
BMS-986397 (CC-91633) is a selective thalidomide-derived molecular glue degrader of CK1α and a modulator of the CRBN E3 ubiquitin ligase. By promoting CRBN-mediated ubiquitination and proteasomal degradation of CK1α, it induces p53 activation and apoptotic pathways in hematopoietic malignancies. BMS-986397 demonstrates potent anti-leukemic activity in acute myeloid leukemia and high-risk myelodysplastic syndrome models, impairing cell proliferation and cell survival.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
