BMH-21 is a small-molecule DNA intercalator that binds ribosomal DNA and inhibits RNA polymerase I (Pol I) transcription. By intercalating into GC-rich genomic regions, BMH-21 disrupts the Pol I transcription machinery and induces proteasomal degradation of RPA194, the catalytic subunit of Pol I. This selective inhibition suppresses ribosome biogenesis, triggers nucleolar stress, and promotes p53-independent apoptosis in cancer cells.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
