Biological Activity

BIX-01294 is a selective inhibitor of the histone methyltransferases G9a and GLP. By specifically reducing cellular histone H3 lysine 9 dimethylation (H3K9me2) levels, BIX-01294 modulates chromatin structure and epigenetic gene repression. This inhibition leads to impaired cancer cell proliferation, induction of autophagy or apoptosis, and suppression of tumor cell growth across various preclinical models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
