Biological Activity

β-Thujaplicin (Hinokitiol) is a natural tropolone derivative and small-molecule iron ionophore that targets transmembrane iron transport mechanisms and downregulates expression of DNMT1 and UHRF1. It acts by transporting iron across lipid bilayers, thereby restoring intracellular iron assimilation and cellular homeostasis. Downstream, β-thujaplicin suppresses Nrf2 signaling pathways, resulting in inhibited cell proliferation, anti-oxidative responses, and anti-tumor activity in biological systems. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
